Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Lyophilised peptide powder is comparatively stable when kept dry, cold and protected from light. Once dissolved, the molecule is exposed to hydrolysis, oxidation and microbial growth, and degradation accelerates at higher temperatures and in alkaline solution. Repeated freeze-thaw cycles concentrate solutes and promote aggregation. Handling guidance for research peptides commonly clusters around freezer temperatures for powder and short refrigerated use for reconstituted solutions, with pH control and sterile technique applied throughout.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.

Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.
Independent verification is central to quality control because the compound is not produced under pharmaceutical manufacturing standards. Third-party laboratories can measure purity, identity, residual solvents, and microbial contamination, though the scope of testing varies between services. Reported analyses of vendor samples have shown batch-to-batch variation in peptide content and the presence of truncated or oxidized species. How much of this variation reflects synthesis conditions versus storage and shipping is not well characterized. No harmonized reference standard exists for the material as sold.
Handling guidance for melanotan II follows general practice for small synthetic peptides rather than a product-specific monograph. Lyophilized powder is typically kept at minus twenty degrees Celsius or colder, protected from light and moisture, because warmth and humidity accelerate degradation. Once reconstituted, solutions are usually refrigerated and used within a short window, as hydrolysis and microbial growth both become concerns. Repeated freeze-thaw cycles are generally avoided. These conventions come from laboratory peptide chemistry and not from formal stability studies on this specific compound.
=== Afonso Dias === In one of the Dias' hearings, it was brought up that the defendant came from a poor family, was 22 and played with an 11 year old child at the time of the disappearance. It was also brought up that the defendant was meeting up with Mendonça in private as Mendonça's parents had forbidden it, after Dias came back changed from the army and as a deserter. Manuel Mendonça said that he and the neighbors noticed that Dias' financial resources increased after the boy's disappearance and that the first PJ team did not believe the information received from both the prostitute and the family. Dias was an unhygienic person, however in the day of the disappearance he took two baths. When Manuel Mendonça got to the PJ they said it was not possible to extract anything from the suspect's clothes. Manuel Mendonça also said that Dias laughed while talking to Filomena Teixeira. He also said that the defendant's lawyer and family did not want to help to find Mendonça. The chief-inspector Henrique Noronha said in 2011 that in the reconstitution there was "a hiatus of time that we couldn't figure out (...) We were a bit perplexed [when Afonso Dias said that he] stood in a place looking at nothing and burning the midnight". The inspector also said that he did not understand the reason of not following the lead of Alcina Dias. Hélder Silva, that was one of the five children that saw Mendonça for the last time, told the Correio da Manhã that Dias "Was very weird. Afonso knew Rui Pedro's whole life, where he was, who he was with, what he would do the next day.
3 ArOH + FeCl3 → Fe(OAr)3 + 3 HCl (Ar = aryl) Among the halide and pseudohalide complexes, fluoro complexes of iron(III) are the most stable, with the colorless [FeF5(H2O)]2− being the most stable in aqueous solution. Chloro complexes are less stable and favor tetrahedral coordination as in FeCl4−; FeBr4− and FeI4− are reduced easily to iron(II). Thiocyanate is a common test for the presence of iron(III) as it forms the blood-red [FeSCN(H2O)5]2+. Like manganese(II), most iron(III) complexes are high-spin, the exceptions being those with ligands that are high in the spectrochemical series such as cyanide. An example of a low-spin iron(III) complex is [Fe(CN)6]3−. Iron shows a great variety of electronic spin states, including every possible spin quantum number value for a d-block element from 0 (diamagnetic) to 5⁄2 (5 unpaired electrons). This value is always half the number of unpaired electrons. Complexes with zero to two unpaired electrons are considered low-spin and those with four or five are considered high-spin. Iron(II) complexes are less stable than iron(III) complexes but the preference for O-donor ligands is less marked, so that for example [Fe(NH3)6]2+ is known while [Fe(NH3)6]3+ is not. They have a tendency to be oxidized to iron(III) but this can be moderated by low pH and the specific ligands used.
Many birds act as ecosystem engineers through the construction of nests, which provide important microhabitats and food for hundreds of species of invertebrates. Nesting seabirds may affect the ecology of islands and surrounding seas, principally through the concentration of large quantities of guano, which may enrich the local soil and the surrounding seas. A wide variety of avian ecology field methods, including counts, nest monitoring, and capturing and marking, are used for researching avian ecology.
While the federal constitution assigns local authorities to the exclusive jurisdiction of state governments, in practice, the federal Ministry of Housing and Local Government oversees the regulation of local laws and policies. Mayors (or presidents for municipal and district councils) and councillors are appointed by the respective state governments, or in the case of the federal territories, by the federal government. Federal laws assign land matters, including the delineation of districts, to the purview of state governments. Except Perlis and the federal territories, each state is divided into districts, which are further subdivided into mukims. In Sabah and Sarawak, districts are grouped into divisions. In contrast to local governments that manage municipal administration and infrastructure development, districts are solely utilised for land taxation. The 13 states are based on historical Malay kingdoms, and 9 of the 11 Peninsular states, known as the Malay states, retain their royal families. The King is elected by and from the nine rulers to serve a five-year term. This King appoints governors serving a four-year term for the states without monarchies, after consultations with the chief minister of that state. Each state has its written constitution. Sabah and Sarawak have considerably more autonomy than the other states, most notably having separate immigration policies and controls, and unique residency status.

channel protein A type of transmembrane protein whose shape forms an aqueous pore in a membrane, permitting the passage of specific solutes, often small ions, across the membrane in either or both directions.
Amplifications (or gene duplications) or repetition of a chromosomal segment or presence of extra piece of a chromosome broken piece of a chromosome may become attached to a homologous or non-homologous chromosome so that some of the genes are present in more than two doses leading to multiple copies of all chromosomal regions, increasing the dosage of the genes located within them. In some cases, only a fraction of a gene is duplicated, producing a gene shorter that the original. In others, the duplicated gene (or fraction of gene) is copied and inserted right after the original DNA sequence (a tandem duplication) resulting in a longer gene. In the long rung, such multiplicative phenomena dominate the distribution of gene lengths. Polyploidy, duplication of entire sets of chromosomes, potentially resulting in a separate breeding population and speciation. Deletions of large chromosomal regions, leading to loss of the genes within those regions. Mutations whose effect is to juxtapose previously separate pieces of DNA, potentially bringing together separate genes to form functionally distinct fusion genes (e.g., bcr-abl). Large scale changes to the structure of chromosomes called chromosomal rearrangement that can lead to a decrease of fitness but also to speciation in isolated, inbred populations. These include: Chromosomal translocations: interchange of genetic parts from nonhomologous chromosomes. Chromosomal inversions: reversing the orientation of a chromosomal segment. Non-homologous chromosomal crossover.
In late November Peter Levene, Chief of Procurement, had a meeting at the Ministry of Defence with his French, German and Italian counterparts (the National Armaments Directors) and the representatives of the consortium, and agreed to "buy European" for certain classes of helicopters, although Heseltine was not actually present. The meeting was later praised by the House of Commons Defence Select Committee. Thatcher, who only learned of the meeting through Cuckney, was displeased, as were Brittan and the Treasury, who thought the US option might be cheaper.
== Methods to screen protein–protein interactions == Though there are many methods to detect protein–protein interactions, the majority of these methods—such as co-immunoprecipitation, fluorescence resonance energy transfer (FRET) and dual polarisation interferometry—are not screening approaches.
Trump is the first president since Grover Cleveland in 1892 to win non-consecutive terms. 2024 also marked the first time since 1892 that the incumbent party had lost in each of three consecutive presidential elections. This was the first time since 1980 that the Democrats were voted out after a single four-year term. This was also the second consecutive election in which the incumbent party had lost after a single four-year term. Trump is the first Republican to win the popular vote since George W. Bush's election in 2004; Bush and Trump are also the only two presidents who won the Electoral College while losing the popular vote in one election who then went on to win both the Electoral College and the popular vote in a subsequent election. Trump is also the first non-incumbent Republican to have won the popular vote since George H. W. Bush in 1988. Trump won a 1.48% margin of victory. While winning the popular vote, Trump did not win a majority of the popular vote; he is the first winning presidential candidate since Bill Clinton in 1996 to win the popular vote with a plurality and also the first Republican since Richard Nixon in 1968 to do so. After Cleveland in 1884 and 1892, Woodrow Wilson in 1912 and 1916, and Clinton in 1992 and 1996, Trump became the fourth president since 1880 to win two presidential elections and receive less than 50% of the popular vote in both presidential election victories. Trump is the first presidential candidate since Barack Obama in 2012 to win a second term in the White House and also the first Republican since George W.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
Mass spectrometry combined with liquid chromatography provides both molecular weight confirmation and a measure of related impurities. Tandem mass spectrometry adds sequence information. A purity percentage reported without a mass measurement does not confirm what the material is.
