Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Solid peptide kept dry at minus twenty degrees Celsius, shielded from light and moisture, is generally considered stable for extended periods. Solutions are divided into single-use aliquots and held at minus twenty or minus eighty degrees Celsius, because repeated freeze-thaw cycles promote aggregation and loss of material to container surfaces. Hydrolysis of the backbone and oxidation of tryptophan are the principal degradation routes in aqueous solution, and both accelerate at ambient temperature. Hygroscopic uptake after a vial is opened can also shift the actual mass weighed, which affects any concentration calculated from it.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.

Melanotan-2 has not received marketing authorisation from major regulatory agencies for any therapeutic indication. Several jurisdictions classify it as a prescription-only medicine or a controlled substance when supplied for human use. Because approved products do not exist, material sold online usually sits outside pharmaceutical supply chains and formal quality oversight. Regulators have issued public notices describing the compound as unapproved. Enforcement varies, and the legal position differs between countries, which complicates any single general statement about its status.
Scientific discussion of Melanotan-2 spans pharmacology, dermatology, and public-health literature. Laboratory studies examine its receptor binding and cellular effects, while clinical reports describe outcomes observed after unregulated use. These two bodies of work differ in rigour and intent. Peer-reviewed trials of the compound as a medicine are limited, so much of the available information comes from case reports and surveillance data. Authors frequently note the gap between experimental findings and real-world use.
Reported observations after unregulated use include shifts in skin pigmentation and, in some accounts, unintended changes to moles and other lesions. Whether these outcomes are causally linked to the compound, and how often they occur, remain open questions because controlled data are scarce. The absence of standardised dosing and verified product purity complicates interpretation. Researchers have called for better surveillance and analytical characterisation of samples obtained outside regulated channels. Conclusions drawn from anecdotal evidence should be treated as provisional.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.
Reversed-phase high-performance liquid chromatography is the routine method for purity assessment. Peptides absorb near 214 nm because of the peptide bond, and a gradient of acetonitrile in water separates the intact peptide from deletion sequences, oxidised products, and earlier-eluting fragments at neutral pH. Electrospray ionisation mass spectrometry provides an orthogonal check: the measured mass must agree with the theoretical value. Amino acid analysis and peptide mapping confirm structure but are used less often. Reference standards remain scarce because the peptide is not described in any pharmacopoeia.
Melanotan-2 is a synthetic cyclic heptapeptide designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous tridecapeptide that regulates pigment production. Two modifications distinguish it from the natural hormone: norleucine replaces methionine at the N-terminus, which limits oxidation, and a D-phenylalanine substitution raises receptor affinity. The ring is closed through an aspartate-lysine lactam bridge, giving the molecule a constrained conformation. The free base has a molecular mass near 1024 daltons, and commercial material is usually supplied as an acetate salt. It appears in the literature as a research peptide rather than an approved therapeutic agent.
Receptor studies place melanotan-2 among non-selective melanocortin agonists, binding MC1R, MC3R, MC4R and MC5R rather than a single subtype. Activation of MC1R on cutaneous melanocytes raises tyrosinase activity and shifts pigment synthesis toward eumelanin, which is darker and more photostable than pheomelanin. Central receptors, particularly MC4R, are associated with appetite suppression and with reported effects on sexual function. Because subtype selectivity is low, the same molecule engages pigment, metabolic and vascular pathways at once, and this breadth is a common explanation offered for the range of adverse events described in user reports.
The Australian Pharmaceutical Formulary (APF) is the national formulary used by pharmacists in Australia, compiled by the Pharmaceutical Society of Australia. New editions of the APF are released every few years, with the latest edition being the 25th.
In the United States Code, the IEEPA is Title 50, §§1701–1707. The IEEPA authorizes the president to declare the existence of an "unusual and extraordinary threat ... to the national security, foreign policy, or economy of the United States" that originates "in whole or substantial part outside the United States." It further authorizes the president, after such a declaration, to block transactions and freeze assets to deal with the threat and requires the president to report to Congress every 6 months on the circumstances, threats and actions taken. In the event of an actual attack on the United States, the president can also confiscate property connected with a country, group, or person that aided in the attack. IEEPA falls under the provisions of the National Emergencies Act (NEA), which means that an emergency declared under the act must be renewed annually to remain in effect. The authority given to the president under the IEEPA does not grant him the ability to regulate or prohibit communication that "does not involve a transfer of anything of value", imports or exports of information or any informational materials, or transactions incidental to travel. Donations intended to relieve human suffering, such as food, clothing or medicine are also excluded unless the president specifically finds their inclusion necessary.
=== Mimicry === Much like the Salticidae family of spiders, P. phalangioides also use mimicry as a predatory tactic to subdue their prey; however, unlike jumping spiders, P. phalangioides do not rely on vision for predation. This mimicry consists of creating specialized vibrations to trick the prey into thinking that it has caught an insect or another spider. The prey then slowly approaches its supposed catch at which point the P. phalangioides spider raises up on its long legs. The spider patiently waits until the exact moment at which the prey touches one of its legs. Then, the P. phalangioides spider quickly immobilizes its prey by using its legs to wrap it up in layers of silk. Its long legs give it plenty of distance from the prey to avoid being bitten in retaliation. After immobilizing its prey, P. phalangioides can administer their venomous bite to the prey and consume it. Even forms of prey that do not fully make it onto the web of P. phalangioides are not safe. Often, prey will trip over the edges of the web, thus providing P. phalangioides with an optimal time to attack. P. phalangioides is capable of clinging onto their web with two of their legs while the rest of their body leans out of the web and shoots silk in the direction of the prey to subdue it.
Nicotinic acid has the formula C6H5NO2 and belongs to the group of the pyridinecarboxylic acids. As the precursor for nicotinamide adenine dinucleotide and nicotinamide adenine dinucleotide phosphate, it is involved in DNA repair. Extra-terrestrial nicotinic acid has been found in carbonaceous chondrite meteorites and in sample-returns from the asteroids 162173 Ryugu and 101955 Bennu.

=== Treatment of deficiency === Severe vitamin B12 deficiency is initially corrected with daily intramuscular injections of 1000 μg of the vitamin, followed by maintenance via monthly injections of the same amount or daily oral dosing of 1000 μg. The oral daily dose far exceeds the vitamin requirement because the normal transporter protein-mediated absorption is absent, leaving only inefficient intestinal passive absorption. Injection side effects include skin rash, itching, chills, fever, hot flushes, nausea and dizziness. There are not enough studies on whether pills are as effective in improving or eliminating symptoms as parenteral treatment.
=== Disease mechanism === In a large proportion (50–70%) of cases, the propensity for malignant hyperthermia is due to a mutation of the ryanodine receptor (type 1), located on the sarcoplasmic reticulum (SR), the organelle within skeletal muscle cells that stores calcium. RYR1 opens in response to conformational changes in the L-type calcium channels following membrane depolarisation, thereby resulting in a drastic increase in intracellular calcium levels and muscle contraction. RYR1 has two sites believed to be important for reacting to changing Ca2+ concentrations: the A-site and the I-site. The A-site is a high affinity Ca2+ binding site that mediates RYR1 opening. The I-site is a lower affinity site that mediates the protein's closing. Caffeine, halothane, and other triggering agents act by drastically increasing the affinity of the A-site for Ca2+ and concomitantly decreasing the affinity of the I-site in mutant proteins. Mg2+ also affect RYR1 activity, causing the protein to close by acting at either the A- or I-sites. In MH mutant proteins, the affinity for Mg2+ at either one of these sites is greatly reduced. The result of these alterations is greatly increased Ca2+ release due to a lowered activation and heightened deactivation threshold. The process of sequestering this excess Ca2+ consumes large amounts of adenosine triphosphate (ATP), the main cellular energy carrier, and generates the excessive heat (hyperthermia) that is the hallmark of the disease.
The end of the war saw the return of American forces in the Philippines. While the Hukbalahap expected to have their war efforts recognized and be treated as allies, the Americans, with the help of USAFFE guerrillas and former PC members, forcibly disarmed Huk squadrons while charging other guerrillas of treason, sedition, and subversive activity, leading to the arrests of Luis Taruc and Casto Alejandrino in 1945, as well as incidents such as the massacre of 109 Huk guerrillas in Malolos, Bulacan. In September 1945, President Sergio Osmeña released Taruc, Alejandrino, and other Huk leaders from prison. The PKP, through Huk leaders, then formally disbanded the movement and formed the Hukbalahap Veterans' League in an effort to get the Hukbalahap recognized as a legitimate guerrilla movement. Alejandrino was its nominal chairman. In 1946, peasants in Central Luzon backed members of the Democratic Alliance in that year's election, with six candidates eventually winning seats in the Senate. Among these candidates was Luis Taruc. However, they were blocked from sitting in Congress by the government, which only exacerbated negative sentiment among the peasants in Central Luzon. The new Roxas administration attempted a pacification program, with help from Taruc, Alejandrino, Juan Feleo, and other representatives. They would be accompanied by MP guards and government officials to try and pacify peasant groups, however this did not result in any sort of success. Within days of the so-called "truce", violence once again erupted in Central Luzon.

Resistin, also known as adipose tissue-specific secretory factor (ADSF) or C/EBP-epsilon-regulated myeloid-specific secreted cysteine-rich protein (XCP1), is a cysteine-rich peptide hormone that is derived from adipose tissue and, in humans, is encoded by the RETN gene. In primates, pigs, and dogs, resistin is secreted primarily by immune and epithelial cells, whereas in rodents, it is mainly secreted by adipose tissue. The human resistin pre-peptide consists of 108 amino acid residues, while in mice and rats it is 114 amino acids in length; the molecular weight is approximately 12.5 kDa. Resistin is classified as an adipose-derived hormone (similar to a cytokine), and its physiological role has been widely debated, particularly regarding its involvement in obesity and type II diabetes mellitus (T2DM).
== Veterinary use == Fentanyl is commonly used for analgesia and as a component of balanced sedation and general anesthesia in small animal patients. The short-acting nature, wide safety profile, lower rate of adverse effect compared to other opioids, and high potency make fentanyl one of the most effective and common analgesics in veterinary medicine; the ability to reduce the minimum alveolar concentration leads to fentanyl being commonly used in dogs that are elderly or have cardiovascular issues. Due to its short duration and quick onset, fentanyl is commonly used as a constant rate infusion or as a transdermal patch, with the latter being able to last 3 to 4 days. Fentanyl can provide sedation for dogs. Fentanyl is less common and has more adverse effects in livestock and equine compared to cats and dogs. In horses at analgesic doses tachycardia, restlessness, and excitement occur. Subcutaneous administration often causes pain, but a bicarbonate solution exists which does not cause pain when given subcutaneously. Fentanyl is highly lipophilic, about 1000 times more than morphine. In dogs it has low protein binding of 15.6% and a half-life of 2–6 hours. In cats the half-life is 2.5 hours. Fentanyl has poor oral bio-availability due to it being metabolized by the liver. Transdermal and transmucosal formulations are common for out-patients, although the latter's efficacy is not well supported by evidence.
=== Other applications === Peptide therapeutics such as are attractive for their high specificity and potency, but they often have poor pharmacokinetic profiles due to their degradation by serum proteases. Though O-GlcNAc is generally associated with intracellular proteins, it has been found that engineered peptide therapeutics modified by O-GlcNAc have enhanced serum stability in a mouse model and have similar structure and activity compared to the respective unmodified peptides. This method has been applied to engineer GLP-1 and PTH peptides.

Purity is normally stated as an area percentage from high-performance liquid chromatography, for example ninety-five or ninety-eight percent. That figure describes the proportion of ultraviolet-absorbing material eluting as the main peak. It says nothing about water content, counterions, residual solvents or mass fraction of the peptide itself.
Peptides are commonly isolated as acetate or trifluoroacetate salts, and the counterion contributes to total mass. Two vials with identical peptide content can therefore differ in weight and in measured response. Residual trifluoroacetate is also relevant in cell-based work because it can influence membrane behaviour.
Degradation products are often closely related to the parent molecule, making them hard to resolve with a single method. Counterion variability adds a further source of scatter between laboratories. Because no pharmacopoeial monograph exists for melanotan-2, groups rely on method-specific validation rather than a shared reference standard.
No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
