The short version of melanocortin fits in a sentence. The long version — which is the one that helps — is below.

Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Two structural changes distinguish the synthetic peptide from the natural hormone. A norleucine residue replaces methionine at one position, and a D-configured phenylalanine replaces the natural L-form at another. Both substitutions slow enzymatic breakdown, which extends the molecule's persistence relative to the parent hormone. The lactam bridge further constrains the backbone into a stable conformation. These features are standard design strategies in peptide chemistry and are not unique to this compound; they appear across many research peptides built for improved stability.
The compound was developed in the late 1980s and early 1990s by academic researchers investigating melanocortin signaling and pigmentation. Early work explored whether synthetic analogs could reproduce effects of the natural hormone under controlled conditions. The molecule never advanced through the full regulatory pathway required for approval as a medicine. From the mid-2000s onward it appeared in unregulated consumer markets, often distributed through informal channels. That gap between research origins and commercial availability shapes how the compound is discussed today.
Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.

==== Effects on mental health ==== Based on a nationally representative sample of 20,847 US adults, a 2026 survey study in JAMA Network Open found that daily generative AI use was significantly associated with greater depressive symptoms, with the odds of at least moderate depression roughly 30% higher among daily users than among non-users. The association was strongest for personal use and among those aged 25 to 64; the cross-sectional design did not allow for causal conclusions. Separately, a 2025 nationally representative survey of US adolescents and young adults (aged 12 to 21) found that roughly 13% had used generative AI for emotional support, with more than 90% of them reporting the advice as helpful. Controlled trials have shown some therapeutic potential. The first randomized controlled trial of a fully generative AI therapy chatbot, published in NEJM AI in 2025, found significant reductions in symptoms of depression, anxiety, and eating disorders over four weeks compared to a waitlist control, with participants rating the therapeutic alliance as comparable to that of human therapists. These findings are consistent with a 2025 meta-analysis of 31 RCTs (n=29,637) that found small-to-moderate effects of AI chatbots in reducing depressive, anxiety, and stress symptoms among young adults.
== Use == The Weimaraner is a versatile hunting dog, and may be used to track, point to, flush or retrieve birds or other game. Registration is subject to successful completion of a working trial. In Germany it is not considered to be suitable for keeping as a companion dog. According to the breed club, it "... basically belongs in the hands of hunters due to its development and its characteristics ... It is not a companion dog, but a hunting dog through and through. As such, it needs work in practical hunting in order to preserve its balanced nature"; whelps are placed mainly with hunters.

The G protein-coupled receptor kinases (GRKs) are protein kinases that phosphorylate only active GPCRs. G-protein-coupled receptor kinases (GRKs) are key modulators of G-protein-coupled receptor (GPCR) signaling. They constitute a family of seven mammalian serine-threonine protein kinases that phosphorylate agonist-bound receptor. GRKs-mediated receptor phosphorylation rapidly initiates profound impairment of receptor signaling and desensitization. Activity of GRKs and subcellular targeting is tightly regulated by interaction with receptor domains, G protein subunits, lipids, anchoring proteins and calcium-sensitive proteins. Phosphorylation of the receptor can have two consequences:
Cells that stop dividing (post-mitotic) and differentiate into neurons early in cortical development are important in laying the groundwork on which other developing neurons can be guided to their proper destination. Tbr1 aids in neuronal migration in the early development of the cerebral cortex. It is largely expressed in post-mitotic neurons of the preplate, which forms a foundation upon which neurons are able to grow and move. As a transcription factor, Tbr1 modulates the expression of RELN, which encodes the Reln protein that forms part of the extracellular matrix of cells. Thus, through regulation of Reln expression, Tbr1 regulates the formation of the matrix through which neurons migrate. Without Tbr1, neurons fail to migrate properly.
Silver and most silver compounds have an oligodynamic effect and are toxic for bacteria, algae, and fungi in vitro. The antibacterial action of silver is dependent on the silver ion. The effectiveness of silver compounds as an antiseptic is based on the ability of the biologically active silver ion (Ag+) to irreversibly damage key enzyme systems in the cell membranes of pathogens. The antibacterial action of silver has long been known to be enhanced by the presence of an electric field. Applying an electric current across silver electrodes enhances antibiotic action at the anode, likely due to the release of silver into the bacterial culture. The antibacterial action of electrodes coated with silver nanostructures is greatly improved in the presence of an electric field. Silver, used as a topical antiseptic, is incorporated by bacteria it kills. Thus dead bacteria may be the source of silver that may kill additional bacteria.
When the B cells get activated, class switching can occur. The class switching involves switch regions that made up of multiple copies of short repeats (GAGCT and TGGGG). These switches occur at the level of rearrangements of the DNA because there is a looping event that chops off the constant regions for IgM and IgD and form the IgG mRNAs. Any continuous looping occurrence will produce IgE or IgA mRNAs. In addition, cytokines are factors that have great effects on class switching of different classes of antibodies. Their interaction with B cells provides the appropriates signals needed for B cells differentiation and eventual class switching occurrence. For example, interleukin-4 induces the rearrangements of heavy chain immunoglobulin genes. That is IL- 4 induces the switching of Cμ to Cγ to Cκ
The latter delivery combination was tested on mice and proved to be effective against leukemia, lymphoma, breast, ovarian, neuroblastoma and prostate cancers. The medium half-life of 227Ac (21.77 years) makes it a very convenient radioactive isotope in modeling the slow vertical mixing of oceanic waters. The associated processes cannot be studied with the required accuracy by direct measurements of current velocities (of the order 50 meters per year). However, evaluation of the concentration depth profiles for different isotopes allows estimating the mixing rates. The physics behind this method is as follows: oceanic waters contain homogeneously dispersed 235U. Its decay product, 231Pa, gradually precipitates to the bottom, so that its concentration first increases with depth and then stays nearly constant. 231Pa decays to 227Ac; however, the concentration of the latter isotope does not follow the 231Pa depth profile, but instead increases toward the sea bottom. This occurs because of the mixing processes, which raise some additional 227Ac from the sea bottom. Thus, analysis of both 231Pa and 227Ac depth profiles allows researchers to model the mixing behavior. There are theoretical predictions that AcHx hydrides (in this case with very high pressure) are a candidate for a near room-temperature superconductor as they have Tc significantly higher than H3S, possibly near 250 K.

There have been a significant number of publications since then, and a Food and Drug Administration panel voted, with some controversy, 20:3 that available studies "supported a signal of harm", but voted 22:1 to keep the drug on the market. The meta-analysis was not supported by an interim analysis of the trial designed to evaluate the issue, and several other reports have failed to conclude the controversy. This weak evidence for adverse effects has reduced the use of rosiglitazone, despite its important and sustained effects on glycemic control. Safety studies are continuing. In contrast, at least one large prospective study, PROactive 05, has shown that pioglitazone may decrease the overall incidence of cardiac events in people with type 2 diabetes who have already had a heart attack.
Most metabolic processes have a specific and narrow range of pH where operation is possible, multiple regulatory systems are in place to maintain homeostasis. Fluctuations away from optimal operating pH can slow or impair reactions and possibly cause damage to cellular structures or proteins. To maintain homeostasis the body may excrete excess acid or base through the urine, via gas exchange in the lungs, or buffer it in the blood. The bicarbonate buffering system of blood plasma effectively holds a steady pH and helps to hold extracellular pH around 7.35. The kidneys are responsible for the majority of acid-base regulation but can excrete urine no lower than a pH of 5. This means that a 330mL can of cola, for example, usually ranging in pH from 2.8 to 3.2, would need to be diluted 100 fold before being excreted. Instead of producing 33L of urine from one can of cola, the body relies on buffer to neutralize the acid. Systemic acidosis can be the result of multiple factors, not just diet. Anaerobic exercise, diabetes, AIDS, aging, menopause, inflammation, infections, tumours, and other wounds and fractures all contribute to acidosis. Blood has an average pH of 7.40 but interstitial fluid can vary. Interstitial pH of the skin, for example, is ~7.1. There is no data available for bone.
where the tangency chord of length k connects the sides of lengths a = e + f and c = g + h, and the one of length l connects the sides of lengths b = f + g and d = h + e. The squared ratio of the tangency chords satisfies
=== Insertion devices === Some pump users prefer to use an insertion device to insert their sets, rather than pushing the needle in by hand. These are spring-loaded and are designed to push the needle into the skin quickly. The user simply loads the primed set into the insertion device, peels off the paper protecting the adhesive pad, places the device against the skin and presses a button. This may be an advantage for those with needle phobia.
=== Nuclear spins and magnets === All nucleons, that is neutrons and protons, composing any atomic nucleus, have the intrinsic quantum property of spin, an intrinsic angular momentum analogous to the classical angular momentum of a spinning sphere. The overall spin of the nucleus is determined by the spin quantum number S. If the numbers of both the protons and neutrons in a given nuclide are even then the system prefers to minimize its spin, giving S = 0, i.e. there is no overall spin. Then, just as electrons pair up in nondegenerate atomic orbitals, so do even numbers of protons or even numbers of neutrons (both of which are also spin-1/2 particles and hence fermions), giving zero overall spin. However, an unpaired proton and unpaired neutron will have a lower energy when their spins are parallel, not anti-parallel. This parallel spin alignment of distinguishable particles does not violate the Pauli exclusion principle. The lowering of energy for parallel spins has to do with the quark structure of these two nucleons. As a result, the spin ground state for the deuteron (the nucleus of deuterium, the 2H isotope of hydrogen), which has only a proton and a neutron, corresponds to a spin value of 1, not of zero. On the other hand, because of the Pauli exclusion principle, the tritium isotope of hydrogen must have a pair of anti-parallel spin neutrons (of total spin zero for the neutron spin-pair), plus a proton of spin 1/2. Therefore, the tritium total nuclear spin value is again 1/2, just like the simpler, abundant hydrogen isotope, 1H nucleus (the proton).

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
It is a synthetic seven-amino-acid peptide modeled on alpha-melanocyte-stimulating hormone. It carries two non-natural substitutions and a cyclic bridge that increase its stability relative to the natural hormone. It circulates as a research chemical and is not an approved medicine.
