This is a working overview of Melanotan II, written for readers who want more than a one-paragraph summary but less than a textbook.

Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.

Of the subjects who were administered Tepezza, 71% in study 1 and 83% in study 2 demonstrated a greater than 2 mm reduction in proptosis (eye protrusion) as compared to 20% and 10% of subjects who received placebo, respectively. The trials were conducted at 28 sites in Europe and United States. Teprotumumab-trbw was approved for use in the United States in January 2020, for the treatment of adults with thyroid eye disease. The FDA granted the application for teprotumumab-trbw fast-track, breakthrough therapy, priority review, and orphan drug designations. The FDA granted the approval of Tepezza to Horizon Therapeutics Ireland DAC.
=== BioCompute and BioCompute Objects === In 2014, the US Food and Drug Administration sponsored a conference held at the National Institutes of Health Bethesda Campus to discuss reproducibility in bioinformatics. Over the next three years, a consortium of stakeholders met regularly to discuss what would become BioCompute paradigm. These stakeholders included representatives from government, industry, and academic entities. Session leaders represented numerous branches of the FDA and NIH Institutes and Centers, non-profit entities including the Human Variome Project and the European Federation for Medical Informatics, and research institutions including Stanford, the New York Genome Center, and the George Washington University. It was decided that the BioCompute paradigm would be in the form of digital 'lab notebooks' which allow for the reproducibility, replication, review, and reuse, of bioinformatics protocols. This was proposed to enable greater continuity within a research group over the course of normal personnel flux while furthering the exchange of ideas between groups. The US FDA funded this work so that information on pipelines would be more transparent and accessible to their regulatory staff. In 2016, the group reconvened at the NIH in Bethesda and discussed the potential for a BioCompute Object, an instance of the BioCompute paradigm. This work was copied as both a "standard trial use" document and a preprint paper uploaded to bioRxiv. The BioCompute object allows for the JSON-ized record to be shared among employees, collaborators, and regulators.
== Later life and recognition == Smolková-Keulemansová became one of the leading experts in the field of chromatography. She was the first professor of chemistry in the Czech Republic and one of the first in Europe. Not only did she continue her studies in chemistry, but she also focused on polarography, a PhD focused on gas chromatography and a DrSc concentrated on inclusion compounds in chromatography. In the early 1970s, inclusion complex formations in selective analytical separations became a major focus of Smolková-Keulemansová's, her first choice being cyclodextrins, but moving on with urea and thiourea for the separation of isomers. Her research on cyclodextrins started soon after her methods focused on gas chromatography, high-performance liquid chromatography and electromigration. Her research became more widespread and she was asked to add many monographs on cyclodextrins, one of them being for a compendium on supramolecular chemistry edited by Jean-Marie Lehn. She has written and co-written 140 original papers and numerous reviews and has contributed to many books, including her work in Journal of High-Resolution Chromatography, "A Few Milestones on the Journey of Chromatography", and an article in the journal Chromatographia, "Study of retention of isomeric aromatic hydrocarbons on GTCB and cyclodextrins". Smolková-Keulemansová died on 27 February 2024, at the age of 96.
As amniotes, reptile eggs have extraembryonic membranes that retain water and facilitate biochemical exchange with the external environment, allowing reptiles to reproduce on dry land, even in extreme, arid habitats. Viviparous species have the eggs developing and hatching inside the mother's body, usually by internal incubation, although some species can nourish the eggs as fetuses through various forms of placenta analogs, with some providing initial parental care for their hatchlings.

A number of structural disorders associated with point mutations of this gene have been described that cause malfunctioning of the heart, such as Type 1R dilated cardiomyopathy and Type 11 hypertrophic cardiomyopathy. Certain defects of the atrial septum have been described recently that could also be related to these mutations. Two cases of dilated cardiomyopathy have been studied involving a substitution of highly conserved amino acids belonging to the protein domains that bind and intersperse with the Z discs. This has led to the theory that the dilation is produced by a defect in the transmission of contractile force in the myocytes. The mutations in ACTC1 are responsible for at least 5% of hypertrophic cardiomyopathies. The existence of a number of point mutations have also been found:
Multiple drugs from each category have been proven effective. Marketized drugs have encountered market demands, also boosted personalized medication developments aiming at a broader range of recipients. Still, disease establishment dilemmas and FDA drug approvals give rise to ethical concerns, posing obstacles in the field's development.
==== New Zealand ==== Off the mainland coast of New Zealand, shelf-edge instability is enhanced in some locations by cold seeps of methane-rich fluids that likewise support chemosynthetic faunas and carbonate concretions. Dominant animals are tubeworms of the family Siboglinidae and bivalves of families Vesicomyidae and Mytilidae (Bathymodiolus). Many of its species appear to be endemic. Deep bottom trawling has severely damaged cold seep communities, and those ecosystems are threatened. Cold seeps are found at depths down to 2,000 m, and the topographic and chemical complexity of the habitats are not yet mapped. The scale of new-species discovery in these poorly-studied or unexplored ecosystems is likely to be high.
=== Storage === Dependent on the storage conditions, garlic powder generally lasts in good condition up to 4 years. Manufactured garlic powders have a 'best before' date printed on them to provide an estimate for how long the powder will be in peak condition for in respect to flavor and texture. Garlic powder must be stored in a cool, dry place, to avoid clumping of the powder. If powder is exposed to moisture or heat, it could cause the product to harden or clump.

It is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, containing seven amino acids with a lactam ring and a D-configured phenylalanine residue. It is supplied as a lyophilised powder for laboratory research.
The natural hormone is a linear tridecapeptide that is rapidly degraded in circulation. Melanotan-2 is shortened, cyclised, N-terminally acetylated, and carries a D-amino acid substitution, all of which slow enzymatic breakdown.
No national medicines regulator has approved melanotan-2 for therapeutic or cosmetic use. In several jurisdictions it is treated as an unapproved prescription medicine, and its legal classification differs from country to country.
No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
