Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
The compound emerged from research programs in the 1980s that examined analogues of alpha-melanocyte-stimulating hormone for pigmentation and photoprotection. Investigators modified the native sequence to extend activity duration and potency. A related analogue, afamelanotide, was developed within the same broad line of inquiry and eventually gained approval in certain jurisdictions for a rare light-sensitivity condition. Melanotan-2 itself did not progress through the same regulatory route and has no approved therapeutic indication.
Lyophilized peptide powder is generally stored frozen, protected from light and moisture. Tryptophan residues are susceptible to oxidation, and the lactam bridge can hydrolyze under strongly acidic or basic conditions. Solutions prepared for laboratory work degrade faster than dry powder, and repeated freeze-thaw cycles accelerate loss. Common practice is to aliquot solutions before freezing and to avoid alkaline buffers. Reported stability windows vary with concentration, buffer, and temperature, so exact shelf lives are method-specific rather than universal.
Regulatory status differs by country, and in many places supplying the compound for human consumption is unlawful. Vendors frequently label material as intended for research use only, a designation that shifts stated purpose but does not create a legal pathway for personal use. Certificates of analysis accompanying such products vary widely in detail and provenance. Third-party testing exists but is voluntary, and results are rarely linked to a specific lot in a publicly verifiable way.

Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.
The peptide was developed during the 1980s by researchers investigating melanocortin signalling and skin pigmentation pathways. Early work focused on analogues of alpha-melanocyte-stimulating hormone that would resist enzymatic breakdown more effectively than the parent molecule. Melanotan-2 emerged from that programme as a shortened, cyclised variant. Reports describing its synthesis and receptor activity later appeared in the scientific literature. Commercial availability grew through unregulated channels rather than through pharmaceutical approval.
Solid peptide kept dry at minus twenty degrees Celsius, shielded from light and moisture, is generally considered stable for extended periods. Solutions are divided into single-use aliquots and held at minus twenty or minus eighty degrees Celsius, because repeated freeze-thaw cycles promote aggregation and loss of material to container surfaces. Hydrolysis of the backbone and oxidation of tryptophan are the principal degradation routes in aqueous solution, and both accelerate at ambient temperature. Hygroscopic uptake after a vial is opened can also shift the actual mass weighed, which affects any concentration calculated from it.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
=== Blue Shift === On February 16, 2021, HECU Collective announced that they would be taking a break from their mod Black Mesa: Azure Sheep and were now focusing on a remake of Half-Life: Blue Shift. Unlike another remake in progress, Guard Duty by Tripmine Studios, the mod utilizes assets from Black Mesa instead of creating it from scratch and is released in chapters. The mod began development after a previous attempt at a Blue Shift remake, Insecurity, was abandoned. Half of the members transitioned to working on the mod while the other half continued work on Azure Sheep. The first of the game's eight chapters was released on March 16, 2021, and as of November 2024, five of the eight total chapters have been released.
Notch is a cell surface protein that functions as a receptor. Animals have a small set of genes that code for signaling proteins that interact specifically with Notch receptors and stimulate a response in cells that express Notch on their surface. Molecules that activate (or, in some cases, inhibit) receptors can be classified as hormones, neurotransmitters, cytokines, and growth factors, in general called receptor ligands. Ligand receptor interactions such as that of the Notch receptor interaction, are known to be the main interactions responsible for cell signaling mechanisms and communication. Notch acts as a receptor for ligands that are expressed on adjacent cells. While some receptors are cell-surface proteins, others are found inside cells. For example, estrogen is a hydrophobic molecule that can pass through the lipid bilayer of the membranes. As part of the endocrine system, intracellular estrogen receptors from a variety of cell types (such as ovary, prostate, lung, cardiovascular and central nervous systems) can be activated by estrogen produced in the ovaries. In the case of Notch-mediated signaling, the signal transduction mechanism can be relatively simple. As shown in Figure 2, the activation of Notch can cause the Notch protein to be altered by a protease. Part of the Notch protein is released from the cell surface membrane and takes part in gene regulation. Cell signaling research involves studying the spatial and temporal dynamics of both receptors and the components of signaling pathways that are activated by receptors in various cell types.
=== La Basilique des Saints-Apôtres (Church of the Holy Apostles) === Alexandre Raymond became interested in Christian art and developed a technique that required great rigour, which we could call “micro-mosaics”. By drawing from the texts written by Procopius of Caesarea, Constantine of Rhodes and Constantine Mazarius, he drew 35 representations of the Church of the Holy Apostles including a series of illustrations of the life of Jesus.
By August, eight of the nine republics, except Ukraine, had approved the draft of the new Treaty with some conditions. Ukraine did not agree on the terms of the Treaty. In the simultaneous Ukrainian republican referendum on 17 March, the majority of residents of Ukraine supported joining the Union on the basis of the 16 July 1990 Declaration of State Sovereignty of Ukraine. In addition, most of autonomous republics expressed the desire to raise their status and to be a party to the new Soviet treaty and declared sovereignty and self-promotion to republics during the parade of sovereignties in the months following the initial proposal from July 1990. Most of them participated in the Novo-Ogaryovo process to draft the treaty, and the status of former autonomous republics was a major point of contention among participants. On July 12, 1991, the Supreme Soviet passed the resolution "About the draft treaty on Union of Sovereign States" to state its stance on the matter that each of "the subjects of the federation, including both the sovereign states - the republics - and the republics incorporated within them on a treaty or constitutional basis" "possesses the right to sign the text of the Union Treaty". As a compromise, the final draft of the treaty allowed a state to join the Union as a part of another state, but only the nine republics were invited to sign it at Novo-Ogaryovo on July 23. Following the August coup, the New Union Treaty was further reformed into the Commonwealth of Independent States.

Smoking 20 cigarettes a day entails a higher risk, but not proportionately. If someone stops smoking, then these chances gradually decrease as the damage to their body is repaired. A year after quitting, the risk of contracting heart disease is half that of a continuing smoker. The health risks of smoking are not uniform across all smokers; risks vary according to the amount of tobacco smoked, with those who smoke more at greater risk. Smoking so-called "light" cigarettes does not reduce the risk.
=== 4 April === The United States government announced a $2.6 billion (USD) aid package to Ukraine which includes ammunition for HIMARS, air defense interceptors, artillery rounds, small arms ammunition, and anti-drone systems.
A Civatte body (named after the French dermatologist Achille Civatte, 1877–1956) is a damaged basal keratinocyte that has undergone apoptosis, and consist largely of keratin intermediate filaments, and are almost invariably covered with immunoglobulins, mainly IgM. Civatte bodies are characteristically found in skin lesions of various dermatoses, particularly lichen planus and discoid lupus erythematosus. They may also be found in graft-versus-host disease, adverse drug reactions, inflammatory keratosis (such as lichenoid actinic keratosis and lichen planus-like keratosis), erythema multiforme, bullous pemphigoid, eczema, lichen planopilaris, febrile neutrophilic dermatosis, toxic epidermal necrolysis, herpes simplex and varicella zoster lesions, dermatitis herpetiformis, porphyria cutanea tarda, sarcoidosis, subcorneal pustular dermatosis, transient acantholytic dermatosis and epidermolytic hyperkeratosis.

In his (August 1962) Report to the National Research Council of the National Academy of Sciences (AP.2), Archie Palmer reported that, of the 945 US "higher education institutions" he had surveyed, 349 had either "formulated and adopted a formalized research and patent policy", or were observing "a generally accepted practice with respect to the handling of research and patent matters", whilst the remaining 596 (which, he observed, conducted "little or no scientific or technological research") had "no formalized research or patent policies". In 1965, 96 US patents were granted to 28 "US universities or related institutions"; in 1992, 1500 US patents were granted to more than 150 "US universities or related institutions" (RH.2, p. 119). In 2025, the (formerly reluctant) University of California had been granted 571 patents, making the university's total "over 6,800 active patents" (JB.1). According to the economists Stanley Engerman and Gavin Wright (SE.1), (a) until 1933 there were more US patents issued to individuals than those issued to corporations, (b) by 1950 the ratio was 50:50, (c) by 1998, whilst the number of US patents issued to individual inventors remained more or less the same, (i.e., 10,000–15,000 per annum), they only represented approx. 10% of the number of patents issued.
Evidence is insufficient to support the use of medications to treat obstructive sleep apnea directly. This includes the use of fluoxetine, paroxetine, acetazolamide, and tryptophan among others. Recent studies are trying to investigate cannabinoids as a treatment for OSA, especially dronabinol, which is a synthetic form of THC (delta-9-tetrahydrocannabinol). Cannabis is known to influence sleep; for example, it can reduce sleep latency. However, results are not consistent. Studies about dronabinol have shown positive impact on the OSA, as they observed a reduced AHI (Apnea-Hypopnea Index) and an increased self-reported sleepiness (the objective sleepiness being unaffected). However, more evidence are needed as many effects of those substances remain unknown, especially the effects of a long-term intake. The effects on sleepiness and weight gain are particularly of concern. Due to uncertainty about its effects and a lack of consistent evidence, medical cannabis is not recommended for the treatment of OSA. There are multiple drugs approved for managing the excessive daytime sleepiness associated with OSA, but not the underlying cause. These include solriamfetol, modafinil, and armodafinil. For adults with obesity and moderate-to-severe OSA, during a 52-week study tirzepatide (brand name Zepbound) substantially reduced apneic-hypopnic events, body weight, hypoxic burden, hsCRP concentration, and systolic blood pressure, and improved sleep-related patient-reported outcomes, all presumably mediated via induced weight loss of 18–20% from baseline.
Deep fascia is a layer of dense fibrous connective tissue which surrounds individual muscles and divides groups of muscles into fascial compartments. This fascia has a high density of elastin fibre that determines its extensibility or resilience. Deep fascia was originally considered to be essentially avascular but later investigations have confirmed a rich presence of thin blood vessels. Deep fascia is also richly supplied with sensory receptors. Histologically, fascia is composed predominantly of type I collagen fibers with variable amounts of elastin, which together determine tensile strength and extensibility. Fibroblasts are the principal resident cells, and fascial tissue contains vascular elements (particularly in deep fascia), immune cells such as macrophages and mast cells, and a dense array of sensory nerve endings; these features enable fascia to participate in repair, inflammation, and nociception. Examples of deep fascia are fascia lata, fascia cruris, brachial fascia, plantar fascia, thoracolumbar fascia and Buck's fascia.
=== Kinetically frozen micelles === When block copolymer micelles do not display the characteristic relaxation processes of surfactant micelles, these are called kinetically frozen micelles. These can be achieved in two ways: when the unimers forming the micelles are not soluble in the solvent of the micelle solution, or if the core forming blocks are glassy at the temperature in which the micelles are found. Kinetically frozen micelles are formed when either of these conditions is met. A special example in which both of these conditions are valid is that of polystyrene-b-poly(ethylene oxide). This block copolymer is characterized by the high hydrophobicity of the core forming block, PS, which causes the unimers to be insoluble in water. Moreover, PS has a high glass transition temperature which is, depending on the molecular weight, higher than room temperature. Thanks to these two characteristics, a water solution of PS-PEO micelles of sufficiently high molecular weight can be considered kinetically frozen. This means that none of the relaxation processes, which would drive the micelle solution towards thermodynamic equilibrium, are possible. Pioneering work on these micelles was done by Adi Eisenberg. It was also shown how the lack of relaxation processes allowed great freedom in the possible morphologies formed. Moreover, the stability against dilution and vast range of morphologies of kinetically frozen micelles make them particularly interesting, for example, for the development of long circulating drug delivery nanoparticles.

No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
Both are synthetic analogues derived from the same parent hormone. Reference descriptions usually characterise afamelanotide as more selective for the MC1R subtype, whereas melanotan-2 is reported to interact with a wider range of melanocortin receptors.
Early studies explored melanocortin signalling and pigmentation, which later led to informal use of the label tanning peptide. That label is not a regulatory category and does not imply approved status for any purpose.
Dry powder is usually held frozen, shielded from light, and kept away from moisture. Desiccant packaging limits hydrolysis during storage. Solutions are typically aliquoted and frozen once, because repeated thawing shortens useful life.
